Canine parvovirus is a highly contagious and potentially fatal viral disease that primarily affects the gastrointestinal tract of dogs. The causative agent, canine parvovirus type 2, is a small, non-enveloped, single-stranded DNA virus belonging to the family Parvoviridae. Since its emergence in the late 1970s as a novel pathogen, canine parvovirus has become one of the most significant infectious diseases of dogs worldwide, particularly threatening unvaccinated puppies and immunocompromised animals.
The virus exists in several antigenic variants, including CPV-2a, CPV-2b, and CPV-2c, which have progressively replaced the original CPV-2 strain through natural evolution. These variants differ in their capsid protein sequences, which affects their antigenic properties and host range. All current variants are capable of causing severe disease in susceptible dogs, and while existing vaccines provide cross-protective immunity against all known variants, ongoing viral evolution necessitates continued surveillance and periodic reassessment of vaccine formulations.
Canine parvovirus targets rapidly dividing cells in the body, which explains its predilection for the intestinal crypt epithelium, lymphoid tissue, and bone marrow. The intestinal tropism results in the characteristic severe enteritis with villous destruction, hemorrhage, and loss of the intestinal mucosal barrier. Concurrent destruction of lymphoid tissue and bone marrow progenitor cells results in profound immunosuppression, which compounds the severity of the disease by predisposing affected dogs to secondary bacterial infections, particularly bacteremia and sepsis.
A myocardial form of parvovirus infection was recognized during the early years of the pandemic, causing acute myocarditis and sudden death in neonatal puppies infected in utero or within the first few weeks of life. This presentation has become exceedingly rare in regions with widespread vaccination, as most breeding bitches now possess adequate antibody levels that provide passive protection to their offspring during the vulnerable neonatal period.
