The MDR1 mutation, also known as the ABCB1 gene mutation, is a genetic variation that affects drug metabolism and transport in certain dog breeds, primarily those with herding heritage. This mutation causes a defect in the P-glycoprotein drug pump, a protein responsible for transporting various substances out of cells, including pumping drugs out of the brain and other sensitive tissues. When this pump does not function properly, certain medications can accumulate to toxic levels in the brain and other organs, causing severe and potentially fatal adverse reactions to drugs that are otherwise safe in dogs without the mutation.
The P-glycoprotein pump functions as a protective barrier, particularly at the blood-brain barrier, where it prevents many drugs and toxins from entering brain tissue. In dogs with the MDR1 mutation, this protective mechanism is compromised, allowing medications to penetrate the brain in much higher concentrations than intended. The result can be severe neurological toxicity including tremors, disorientation, blindness, seizures, coma, and death. Understanding this mutation is crucial for any owner of an affected breed or mixed breed dog, as awareness can literally save lives by preventing exposure to dangerous medications.
The MDR1 mutation is inherited in an autosomal incomplete dominant pattern, meaning dogs can be normal (two normal copies of the gene), carriers or heterozygous (one mutant and one normal copy), or homozygous affected (two mutant copies). Dogs that are homozygous affected are most sensitive to problematic medications and can experience severe reactions even at normal therapeutic doses. Heterozygous dogs have intermediate sensitivity and may react to higher doses or certain particularly problematic drugs. Approximately 75 percent of Collies carry at least one copy of the MDR1 mutation, with similar high prevalence in several other herding breeds.
Genetic testing for the MDR1 mutation is readily available, relatively inexpensive, and strongly recommended for all dogs of affected breeds or those with herding breed ancestry. Testing requires only a simple cheek swab that can be collected at home and mailed to a laboratory, with results typically available within one to two weeks. Knowing a dog's MDR1 status allows veterinarians to avoid problematic medications or adjust doses appropriately, preventing potentially fatal adverse reactions. All owners of herding breeds should have their dogs tested before any medications are prescribed, and this information should be prominently noted in the dog's medical records.
