As the active metabolite of toltrazuril, ponazuril shares the same potent antiprotozoal activity while being more readily available in some markets. Unlike coccidiostatic medications that merely inhibit parasite growth, ponazuril acts as a true coccidiocide, directly killing coccidia parasites and often achieving complete parasite elimination with short treatment courses. This makes ponazuril particularly valuable when rapid and thorough resolution of coccidial infections is desired.
The mechanism of action of ponazuril involves disruption of multiple stages of the coccidial life cycle through interference with the parasite's intracellular development and reproductive processes. The drug damages coccidia during both asexual stages (schizonts and merozoites) and sexual stages (gametocytes), preventing the formation of viable oocysts that would otherwise be shed in feces to infect other animals. Research has demonstrated that ponazuril affects parasite cellular structures including the outer membrane, causes marginalization of chromatin, and disrupts mitochondrial function within the parasites. Ponazuril is available primarily as the Marquis oral paste formulated for horses, which contains 15% ponazuril. For rabbit use, veterinarians typically prescribe compounded oral suspensions that provide more appropriate concentrations for the smaller doses required. Compounding pharmacies can prepare ponazuril in liquid formulations at concentrations suitable for accurate dosing in rabbits. The medication is generally well-tolerated, and studies in rabbits have demonstrated an excellent safety profile even at doses higher than those typically used for treatment. Treatment protocols are often short, with research showing complete parasite eradication possible with as few as three days of treatment.
Veterinary guidance is essential when using ponazuril in rabbits because the medication requires proper compounding for appropriate dosing and is used in an extra-label manner. A rabbit-savvy veterinarian can diagnose coccidiosis through fecal examination, determine the severity of infection, prescribe the appropriate formulation and dose, and monitor treatment response.
