While healthy dogs typically obtain sufficient vitamin K1 from their diet and bacterial synthesis in the intestines, therapeutic supplementation becomes essential in certain clinical situations, most notably anticoagulant rodenticide poisoning.
The mechanism of action of vitamin K1 involves its role as a cofactor for the enzyme vitamin K epoxide reductase complex, which is essential for the post-translational modification of certain clotting factor precursors. Specifically, vitamin K1 enables the carboxylation of glutamic acid residues on clotting factors, a chemical modification absolutely required for these proteins to bind calcium and participate in the coagulation cascade. When vitamin K1 is depleted or its recycling is blocked, as occurs with anticoagulant rodenticide poisoning, the liver produces inactive clotting factor precursors, and normal clotting becomes impossible. Administering vitamin K1 restores the ability to produce functional clotting factors.
Vitamin K1 is available in several formulations for veterinary use including tablets, capsules, oral liquid preparations, and injectable solutions. Oral formulations are most commonly used for ongoing treatment and prevention, while injectable forms may be used in emergency situations when rapid effect is needed or when oral administration is not possible. The injectable form can be given subcutaneously or intramuscularly, with subcutaneous administration generally preferred due to better absorption compared to intramuscular injection. Intravenous administration is typically avoided due to the risk of anaphylactic reactions.
Vitamin K1 is critically important in veterinary medicine due to the prevalence of anticoagulant rodenticide poisoning in dogs. These rodent poisons work by blocking vitamin K1 recycling in the liver, eventually depleting the body's vitamin K1 stores and causing fatal bleeding. Second-generation anticoagulant rodenticides such as brodifacoum and bromadiolone are particularly dangerous because they persist in the body for weeks to months, requiring extended vitamin K1 therapy. Prompt recognition and treatment of rodenticide poisoning with vitamin K1 can be lifesaving, making this vitamin one of the most important therapeutic agents in veterinary emergency medicine.
