Chocolate contains two closely related methylxanthine compounds that are toxic to dogs: theobromine and caffeine. While humans metabolize these substances relatively quickly, dogs process them far more slowly, allowing the compounds to accumulate to toxic levels in the body. Theobromine is the primary toxicant of concern, as it is present in chocolate in much higher concentrations than caffeine. The half-life of theobromine in dogs is approximately 17.5 hours, meaning it takes nearly three-quarters of a day for the body to eliminate just half of the absorbed dose.
Theobromine and caffeine exert their toxic effects through multiple mechanisms. They act as competitive antagonists of adenosine receptors, which normally promote relaxation and reduce neural activity. By blocking adenosine, methylxanthines stimulate the central nervous system and cardiovascular system, leading to restlessness, increased heart rate, and heightened neural excitability. They also inhibit phosphodiesterase enzymes, resulting in elevated intracellular cyclic AMP levels that amplify cellular signaling and contribute to cardiac stimulation.
Additionally, methylxanthines increase intracellular calcium concentrations in cardiac and skeletal muscle cells, enhancing contractility but also increasing the risk of arrhythmias at toxic doses. They promote catecholamine release, further stimulating the heart and blood vessels. At the kidney level, theobromine acts as a diuretic by inhibiting tubular reabsorption of sodium and water, contributing to increased urination and potential dehydration in poisoned dogs.
The toxic dose of theobromine varies by individual dog, but clinical signs generally begin to appear at doses of approximately 20 milligrams per kilogram of body weight. Cardiac effects such as tachycardia and arrhythmias typically manifest at doses above 40 to 60 milligrams per kilogram. Seizures and potentially fatal complications can occur at doses exceeding 60 milligrams per kilogram, although lethal doses have been reported in the range of 100 to 200 milligrams per kilogram. Because the toxic dose depends on the dog's size, small dogs are at disproportionately greater risk from any given amount of chocolate.
