The mechanism of action of permethrin involves disruption of nerve impulse transmission in arthropods through modification of voltage-gated sodium channel function. Upon contact with target parasites, permethrin molecules bind to sodium channels in nerve membranes, preventing normal channel inactivation and causing prolonged sodium influx. This results in repetitive nerve firing, hyperexcitation of the nervous system, and ultimately paralysis and death of the affected insect or arachnid. The rapid knockdown effect characteristic of permethrin and other pyrethroids provides quick incapacitation of parasites, limiting the duration of host contact and associated blood feeding or tissue damage before pest mortality occurs.
Permethrin ear tags are manufactured as polymer matrices containing the active ingredient distributed throughout the tag material for extended-release delivery over three to five months. The insecticide gradually migrates from the interior of the tag to the surface, where it transfers to the hair coat through volatilization, direct contact during head movements, and animal-to-animal contact during social interactions and grooming. This distribution mechanism creates a protective layer of insecticide that extends across the entire body from the localized ear placement, providing comprehensive coverage against mobile ectoparasites. The physical properties of permethrin, including moderate vapor pressure and appropriate lipophilicity, optimize distribution through the sebaceous coat.
Permethrin ear tags are regulated by the Environmental Protection Agency and are available over the counter without veterinary prescription for use in beef cattle and non-lactating dairy cattle. The compound's favorable mammalian safety profile results from efficient metabolic degradation in mammalian systems compared to insects, creating a wide margin between effective insecticidal doses and toxic thresholds in cattle. However, decades of intensive permethrin use have contributed to widespread resistance development in horn fly populations, necessitating careful attention to resistance management principles including rotation with alternative chemical classes and adherence to best practices for application timing and tag removal.
