Vitamin K is a fat-soluble vitamin essential for the synthesis of blood clotting factors, playing a critical role in hemostasis and preventing uncontrolled bleeding in guinea pigs and all mammals. The vitamin K family includes phylloquinone (vitamin K1), which is obtained from plant sources, and menaquinones (vitamin K2), which are produced by bacteria including those in the gastrointestinal tract. Under normal circumstances, guinea pigs obtain adequate vitamin K from their diet of hay and vegetables combined with vitamin K synthesized by gut bacteria and absorbed through cecotrophy, the consumption of nutrient-rich cecal pellets.
The mechanism of action of vitamin K centers on its essential role as a cofactor for the enzyme gamma-glutamyl carboxylase, which activates several proteins required for blood coagulation. These vitamin K-dependent clotting factors include factors II (prothrombin), VII, IX, and X, as well as anticoagulant proteins C and S. Without adequate vitamin K, these clotting factors cannot undergo the carboxylation reaction necessary for their function, leading to impaired coagulation and increased bleeding risk. This biochemical pathway explains why vitamin K deficiency or antagonism results in dangerous bleeding disorders.
Vitamin K supplementation in guinea pigs is most commonly required in emergency situations involving rodenticide poisoning, as many rodent poisons work by blocking vitamin K recycling and thereby preventing blood clot formation. These anticoagulant rodenticides, including warfarin and newer long-acting compounds, cause progressive depletion of vitamin K-dependent clotting factors, leading to potentially fatal internal and external bleeding. Vitamin K1 (phylloquinone) serves as the specific antidote for anticoagulant rodenticide poisoning and must be administered promptly and continuously until the poison is cleared from the body.
The safety profile of vitamin K is generally excellent, as the vitamin is not toxic even at relatively high doses when administered appropriately. Unlike vitamins A and D, which can cause toxicity with excessive intake, vitamin K does not accumulate to harmful levels under normal circumstances. Injectable vitamin K1 (phytonadione) is the most commonly used form in veterinary emergency medicine for treating rodenticide poisoning. Oral vitamin K supplements may be used for ongoing treatment or for addressing less severe deficiency states. An exotic veterinarian should oversee vitamin K therapy, as proper dosing and duration are critical for successful treatment of poisoning cases.
