The mechanism of action of L-asparaginase involves depletion of asparagine, an amino acid required for protein synthesis. While normal cells can produce their own asparagine through the enzyme asparagine synthetase, many lymphoid cancer cells lack this capability and depend entirely on circulating asparagine from the bloodstream. When L-asparaginase is administered, it rapidly breaks down asparagine in the blood and extracellular fluid, essentially starving cancer cells that cannot manufacture their own. This results in inhibition of protein synthesis, cell cycle arrest, and ultimately death of the asparagine-dependent malignant cells.
L-asparaginase is available exclusively as an injectable formulation and must be administered by a veterinarian. The medication is typically given by intramuscular, subcutaneous, or intravenous injection, depending on the protocol being used and the preference of the treating veterinarian. Because L-asparaginase is a protein-based medication derived from bacterial sources, there is an inherent risk of allergic reactions, and administration in a veterinary setting where emergency treatment is available is essential. The drug is not suitable for home administration by owners due to both the injection requirement and the need for monitoring during and after treatment.
The use of L-asparaginase in guinea pigs requires careful veterinary supervision, particularly regarding the risk of hypersensitivity reactions which can range from mild to life-threatening. Guinea pigs, like all patients receiving this medication, may develop antibodies to the bacterial enzyme over time, increasing the risk of allergic reactions with repeated doses.
