Megacolon is a serious genetic condition in rabbits characterized by an abnormally enlarged and poorly functioning colon. This hereditary disorder is directly linked to the En (English spotting) gene, which controls the broken coat pattern seen in breeds like the English Spot, Checkered Giant, and Rhinelander. When a rabbit inherits two copies of the En gene (homozygous), the result is not only an almost entirely white coat with minimal markings but also a malformed enteric nervous system that prevents normal intestinal function. These double-En rabbits, often called "charlies" due to their distinctive appearance, face lifelong digestive challenges that require dedicated management.
The condition develops because the En gene affects neural crest cell migration during embryonic development. Neural crest cells are responsible for forming the enteric nervous system, which controls the rhythmic contractions (peristalsis) that move food through the digestive tract. In megacolon rabbits, portions of the colon lack adequate nerve supply, leading to poor motility, chronic constipation, and progressive dilation of the affected intestinal segments. This creates a cycle where the colon stretches further as fecal material accumulates, worsening the underlying dysfunction.
Megacolon significantly impacts affected rabbits' quality of life and requires substantial owner commitment. Without proper management, these rabbits experience chronic discomfort, recurrent GI stasis episodes, and potentially fatal impactions. The condition cannot be cured, as it stems from a fundamental structural abnormality present from birth. However, with appropriate dietary modifications, regular monitoring, and prompt veterinary intervention when problems arise, many megacolon rabbits can live reasonably comfortable lives, though their lifespan may be shortened compared to unaffected rabbits.

Responsible breeders avoid breeding two broken-patterned rabbits together specifically to prevent producing charlie offspring.
