Cottontail Rabbit Papillomavirus, also known as Shope Papilloma Virus or CRPV, is a viral disease caused by a DNA virus of the Papillomaviridae family that affects rabbits and certain hares. The virus was first discovered and characterized by Richard E. Shope in 1933 when investigating unusual horn-like growths on wild cottontail rabbits in the Midwestern United States, making it one of the first viruses identified as causing tumors in mammals. This groundbreaking discovery contributed significantly to the understanding of viral oncogenesis and later facilitated the development of human papillomavirus vaccines. The natural host for CRPV is the eastern cottontail rabbit, but the virus can also infect domestic European rabbits, brush rabbits, jackrabbits, and snowshoe hares.
The virus causes characteristic wart-like growths called papillomas that typically appear on hairless or thinly-haired skin areas including the ears, eyelids, lips, nose, and occasionally the neck, shoulders, and abdomen. In cottontail rabbits, these lesions may appear as rough, raised, reddish growths that can develop into distinctive horn-like keratinized structures over time. The virus is transmitted primarily through biting insects including mosquitoes, ticks, and other arthropods that carry viral particles from infected to susceptible rabbits. Direct contact transmission is also suspected though less well documented.
The significance of CRPV infection lies primarily in its oncogenic potential, particularly in domestic rabbits. While the virus naturally coexists with cottontail rabbits and causes largely benign infections in that species, domestic European rabbits are more susceptible to malignant transformation. In domestic rabbits, papillomas that persist beyond approximately six months have a substantial risk of progressing to squamous cell carcinoma, with malignant transformation occurring in sixty-six to eighty percent of persistently infected domestic rabbits compared to approximately twenty-five percent of cottontails. These carcinomas can metastasize to lymph nodes, lungs, and other organs, representing a potentially life-threatening condition.
Treatment approaches for CRPV infection include surgical removal of papillomas before malignant transformation occurs, supportive care, and monitoring for regression or progression. Many papillomas regress spontaneously within six months as the immune system mounts an effective response, making observation a reasonable initial approach for small, non-problematic lesions. However, domestic rabbit owners should be aware of the significant cancer risk associated with persistent lesions and work closely with a rabbit-savvy veterinarian to monitor for progression and determine appropriate intervention timing. Prevention focuses on minimizing exposure to insect vectors and avoiding contact with infected wild rabbits.
